i saved my single product image gallery as in the screen shot and save. but its not happen as like the second ss

Maybe you are using Custom WooCommerce Builder layouts, which are part of WooCommerce Builder from WoodMart.
Navigate to Layouts > Edit Single Product Layout and in the layout, edit the product gallery and choose the gallery layout.

if i delete the single product page from this layout? what will happen ?

If you delete the Single Product layout from the Layout Builder, your site will fall back to the default WooCommerce product layout provided by the theme. If you want to use the default design, you can disable the layout.
